Autophagy in Chronic Inflammation during Aging- EISENBERG Tobias (MetAGE Key Researcher)

Background:

Aging is associated with chronic (sterile) inflammation and immunosenescence, processes that contribute significantly to age-related diseases. Autophagy, a cellular recycling pathway, plays a vital role in maintaining proteostasis and organelle homeostasis, ensuring proper immune cell function in older adults. Consequently, changes in autophagic activity are hypothesized to drive both immunosenescence and chronic inflammation. Irrespectively, the exact mechanisms by which aging and aging-associated conditions, including obesity, metabolic syndrome, and cardiometabolic dysfunction, influence autophagy in humans remain poorly understood. This gap in knowledge is partly due to past methodological limitations.

Hypothesis and Objectives:

We hypothesize that chronic inflammation derives from cell type-specific alterations in autophagy in an age-dependent manner, contributing to a vicious cycle between inflammation and autophagic dysfunction. By applying a novel method to assess autophagic flux, developed within the MetAGE framework, along with advanced single-cell technologies, this project aims to elucidate how autophagy is regulated in circulating immune cells in humans and animal models during aging and in response to geroprotective interventions. Additionally, we seek to understand how these autophagic changes are linked to chronic (sterile) inflammation.

Methodology:

As part of the MetAGE Cluster of Excellence, you will investigate the interplay between inflammation and autophagy using various assays, including autophagic flux measurements with flow cytometry. You will complement this with biochemical and omics analyses to explore inflammatory signaling pathways. Collaboration with the MetAGE consortium will provide access to both animal models and human samples from controlled trials and observational studies, enabling translation of preclinical findings into human aging research.

We offer an annual gross salary of € 52,007.20 for a fulltime position.
